Interactions of Quaternary Ammonium Compounds and Proteins

چکیده
A simple met.hod of titration with a cationic detergent has been developed which permits rapid estimation of the concentration of urinary protein with a minimum of manipulations and time. The method depends on the formation of an insoluble anion-cation complex between quaternary ammonium ions, hereinafter referred to as the cationic detergent, and proteins, in the present instance mainly human albumin, at alkaline pH. No protein is precipitated from solution by the addition of the cationic detergent until a slight excess of this is present, at which time the solution shows faint but definite turbidity. Addition of more cationic detergent results at first in increasing turbidity, but as the addition is continued the turbidity decreases, almost as abruptly as it appeared, to give a nearly clear solution. Further addition of the cationic detergent is not attended by reappearance of turbidity. The end-point used in the present method is the appearance of a definite turbidity. The protein-quaternary ammonium ion complex formation is sensitive to ionic strength, non-electrolyte concentration, pH, and type of protein present (see, for example, Polonovski and Macheboeuf (l), Schmidt (2), and Valko (3)). For this reason, the method is restricted for accurate results to urine protein of the order of 2 or more gm. per liter (0.4 or more mg. of protein per sample) in order that the dilutions made may be sufficient to reduce the factors of urine ionic strength, non-electrolytes, and pH to negligible proportions. The method is, however, applicable to most cases of significant proteinuria. The simplicity of the procedure is indicated by the fact that we have been able to complete nearly 200 analyses in an afternoon. If the end-point is determined by the optical density change in a photometer, the method can be used for samples containing as little as 0.02 mg. of protein.

Use of green fluorescent protein labeled non-tuberculous mycobacteria to evaluate the activity quaternary ammonium compound disinfectants and antibiotics
Although infections with NonTuberculous Mycobacteria have become less common in AIDS patients, they are important opportunistic infections after surgical procedures, likely because they are ubiquitous and not efficiently killed by many commonly used disinfectants. In Venezuela there have recently been many non-tuberculous mycobacteria soft tissue infections after minor surgical procedures, some...
متن کاملRole of the ammonium group in the diffusion of quaternary ammonium compounds in Streptococcus mutans biofilms.
OBJECTIVES Cetylpyridinium chloride (CPC), a quaternary ammonium compound, was shown to interact irreversibly with Streptococcus mutans biofilms, leading to a slow diffusion compared with poly(ethylene glycol) (PEG) molecules of similar size. The objective of this work is to determine if the retardation of CPC diffusion and its strong binding to biofilms is caused by interactions between the am...
متن کاملGenetic basis of resistance to quaternary ammonium compounds – the qac genes and their role: a review
Although the qac genes are named after one of their main substrates (i.e., quaternary ammonium compounds), these genes also code for resistance to a broad spectrum of other cationic compounds such as intercalating dyes, diamidines and biguanides. The various Qac proteins are involved in relatively low specific effluxbased multidrug pumps and belong to a family of small multidrug resistance prot...
متن کاملCharacterization of the quaternary amine transporters of Rhizobium leguminosarum bv. viciae 3841.
Rhizobium leguminosarum bv. viciae 3841 contains six putative quaternary ammonium transporters (Qat), of the ABC family. Qat6 was strongly induced by hyperosmosis although the solute transported was not identified. All six systems were induced by the quaternary amines choline and glycine betaine. It was confirmed by microarray analysis of the genome that pRL100079-83 (qat6) is the most strongly...

Bacterial cellulose (BC) has attracted attention for preparing advanced materials due to its microfibrous structure. In this study, quaternary ammonium bacterial cellulose (QABC) as well as quaternary ammonium plant cellulose (QAPC) as adsorbents for proteins have been prepared. Introduction of the quaternary ammonium group was conducted under different conditions to determine the best conditio...
